The chancellor is the chief academic and administrative officer for the University of Colorado Denver. She reports to the CU system president for the conduct and affairs of CU Denver.

Michelle Marks became CU Denver’s chancellor on July 1, 2020. A noted innovator in higher education, Chancellor Marks spent her first 100 days gathering input to assess CU Denver’s progress and priorities. In January 2021, she launched a strategic planning process to chart a course for CU Denver's future.
Vice Chancellors and Cabinet Members support the Chancellor by overseeing various aspects of university administration.
The Provost and Executive Vice Chancellor oversees the academic, faculty, and student programs at the University of Colorado Denver and collaborates with Anschutz Medical campus on academic programming and accreditation.
The Deans are responsible for academic operations and administration of their respective schools and colleges, and report to the Provost.
