Genetic and Environmental Determinants of Pulmonary Fibrosis

Project Description

Project 1 The overall goals of this proposal are to comprehensively identify the specific sequence variants involved in the development of idiopathic pulmonary fibrosis (IPF), to explore how these key IPF sequence variants contribute to the etiologic, biologic, and clinical phenotypes of this disease, and then examine the generalizability of these genetic risk variants to other ethnic groups.    Project 2 The overall goal of the proposed research is use a systems biology approach to develop an integrated understanding of how sequence variants and epigenetic marks contribute to the development of idiopathic pulmonary fibrosis (IPF).    Project 3 The overall goals of this proposal are to advance a therapeutic intervention for preclinical or mild forms of IPF by targeting the glycoprotein MUC5B with a novel mucolytic agent and to develop a biomarker profile relevant to the detection of preclinical/mild IPF.    Project 4 The overall goal of this proposal is to understand how dysregulated MUC5B results in the development of fibroproliferative lung disease. We have engineered an efficient, tiered approach to understand how dysregulated MUC5B results in the development of fibroproliferative lung disease.  Based on what is known about IPF and MUC5B, we hypothesize that chronic mucus hypersecretion impairs mucociliary transport and results in persistent cell injury causing impaired host defense and excessive repair and leading to the development of fibroproliferative lung disease.
